PNC-27 pre-mixed pen contains a synthetic peptide that selectively kills cancer cells, making it potentially useful as a cancer treatment. However, PNC-27 is still a research peptide and has not yet been cleared by the FDA for human use.
According to research, the PNC-27 peptide may attach to receptors on cancer cells, inducing cell death. It is a component of the PNC sequence of research proteins designed to mend and cause cell necrosis in deformed (cancerous) cells while leaving normal, healthy cells uninjured and intact.
The PNC-27 peptide has a bound HDM-2 domain and a transmembrane-penetrating domain, which correspond to p53 residues 12–26. This has been shown to allow the peptide to bind and kill cancer cells by membranolysis or cell membrane breakdown.

In a 2009 study, researchers revealed that persons who ingested pnc 27 could see the peptide’s p53 residues superimposed on the structure of hdm2. As a result, PNC-27 can penetrate the membranes of hdm-2 cancer cells [1].
Because cancer cells contain a high concentration of HDM in their membranes, the PNC-27 pre-mixed pen can selectively target malignant cells while leaving healthy cells alone.
Scientists also discovered that implanting hdm2 into untransformed cells that were previously resistant to PNC-27 made them susceptible to the peptide. These findings demonstrate that the PNC-27 peptide can target HDM-2 in cancer cell membranes and destroy them by membranolysis [2].
Meanwhile, another study published in 2010 by trained scientists revealed that PNC-27 attacked cancer cells completely rather than in pieces [3]. The researchers wanted to know whether the pores in the cancer cell’s membrane were produced by individual peptides or the full peptide. The study determined that it was the latter.
The researchers observed that the whole peptide was responsible for the effects on the cancer cell membrane, whereas the surrounding noncancerous cells were unaffected.
The PNC-27 pre-mixed pen has a variety of practical benefits, including pain relief within one week of receiving the peptide. Within three weeks, animal participants develop flu-like symptoms, indicating that their immune systems are responding to the destruction of cancer cells. Furthermore, bilirubin and lactate dehydrogenase levels rise after six weeks.
Significant tumour breakdown and softening does not occur for ten weeks. In some cases, the tumour site may enlarge. However, this is associated with the immune system’s inflammatory response.
After three months, scientists report that animal participants have less cancer-related symptoms and have more vitality.
