Description
Ipamorelin Nasal Spray
Ipamorelin has five amino acids in its structure and is known as a pentapeptide. It is a GH secretagogue and an agonist, meaning it may bind to specific cell receptors and cause a physiological reaction. The peptide’s mechanism of action allows it to boost pituitary gland-based growth secretion expression in animal test participants. Moreover, the peptide has been found to reduce the production of somatostatin. Ipamorelin can also increase IGF-1, or Insulin-like Growth Factor 1. It is essential for the growth and repair of muscle and skeletal tissue.
Ipamorelin is a peptide that binds to the ghrelin/growth hormone secretagogue receptor. A study found that it had no effect on ACTH, prolactin, FSH, LH, TSH, or cortisol secretion [1]. It is one of the most selective GH secretagogues known. Because of its great specificity, ipamorelin has been studied as a treatment as well as a model peptide for learning how selectivity in receptor binding is produced.

Research
One of the most prevalent adverse corticosteroid effects is Ipamorelin, which has been shown to reduce inflammation in diseases ranging from cancer to autoimmune illness. Mitigating glucocorticoid side effects may allow higher doses and more prolonged use of these drugs, reducing morbidity and mortality. Several studies have indicated that ipamorelin nasal spray can minimize or reverse glucocorticoid adverse effects.
Bone Health:
Long-term glucocorticoid use causes bone loss and an increased risk of fracture. Bisphosphonates, hormones, and novel monoclonal antibodies are current treatments. These treatments work, but they have adverse effects, are ineffective, or are expensive. However, ipamorelin nasal spray is relatively cheap to make and has few adverse effects. In rodent studies, ipamorelin nasal spray reverses corticosteroid-induced bone loss and increases bone growth fourfold [2].
Further research shows that ipamorelin enhances bone mineral density and thus the strength of existing and freshly created bone. Also, ipamorelin counteracts some of the adverse effects of steroids, like muscle loss and higher visceral fat deposition [3].
Muscle Growth:
GH and growth hormone secretagogues like ipamorelin have been shown to attenuate the muscle catabolic effects of glucocorticoids. Ipamorelin reduces liver nitrogen wasting and improves nitrogen balance in glucocorticoid-treated rats. Muscle wasting is a common treatment-limiting adverse effect of glucocorticoids. For patients on glucocorticoids, the potential to reverse muscle catabolism and bone loss with a single medication could be enormously advantageous [2].
Inhibition of ghrelin receptor signalling by Ipamorelin
The ghrelin receptor is known to be overexpressed in cancer (e.g. human carcinomas) and heart failure patients. This is why scientists have proposed using ipamorelin as a PET probe to diagnose cancer. A PET probe made of ipamorelin, which is easily synthesized in the lab, has been shown to be feasible in primary in vitro experiments. This will be followed by in vivo testing and criteria for interpreting PET tests performed with the probe [4].
Diabetes:
Ipamorelin nasal spray can increase insulin release in diabetic rats. This impact is probably due to indirect calcium channel stimulation in pancreatic islet cells that make and store insulin. The effects of ipamorelin on the pancreas may help us comprehend the constraints of type 2 diabetes and develop new therapies or even prevention approaches [5].
Post-operative ileus (POI) is a common complication of surgery, particularly abdominal surgery. The Gl system malfunctions, preventing individuals from taking oral nutrients. Although it causes discomfort, POI slows hospital discharge and overall recovery. Several proof-of-concept clinical trials have looked at ipamorelin nasal spray to see if it can lessen POI.
Ipamorelin nasal spray appears to reduce the time to about 12 hours. Despite early moderate success, the trials’ business considered efficacy was not high enough to generate a commercial product. Ongoing research may improve efficacy or discover a synergistic effect between ipamorelin and other drugs, making combination therapies more effective [6].
Summary
Despite not being an orphan drug, Ipamorelin is a neglected medication in research settings. Despite encouraging early research, ipamorelin’s popularity faded after being abandoned as a post-operative ileus therapy.
Nevertheless, Ipamorelin nasal spray has many uses, not only as a medicinal but also as a tool for better understanding illness states and their physiologic effects. Ipamorelin nasal spray will likely be researched again once additional data and cutting-edge insights into the impact of this unique peptide are released.
